Bacterial Media



Luria Broth (LB)

To make 1L LB media:

  • 10 g Bacto-tryptone
  • 5 g NaCl
  • 5 g Yeast extract
Dissolve in a final volume of 1L. Pour into 100 ml bottles and autoclave.

To make 1L LB plates:
  • 10 g Bacto-tryptone
  • 5 g NaCl
  • 5 g Yeast extract
  • 15 g Agar
Dissolve in a final volume of 1L and autoclave.

To make LBamp plates:
Allow LB medium to cool to ~50oC after autoclaving, and add 1 ml of a 100mg/ml ampicillin stock.

SOB media

To make 1L SOB media:

  • 20 g Bacto-tryptone
  • 0.5 g NaCl
  • 5 g Yeast extract
  • 900 ml distilled H2O
  1. Shake until the solution dissolves.

  2. Add 10 ml 250mM KCl.

  3. Adjust pH to 7.0 with 10N NaOH.

  4. Bring to a final volume of 1L with H2O. Pour into 100 ml bottles and autoclave.

  5. To each bottle, add 500 ul 2M MgCl2.

SOC media

To make 1L SOC media:

  1. Prepare SOB media as above.

  2. Before adding the MgCl2, add 1.8ml 2% glucose to each 100ml bottle.

  3. To each bottle, add 500 ul 2M MgCl2.
